Abstract

Understanding soil microbial diversity is essential for ecosystem stability, particularly under the duress of climate change. This study employs a comparative framework to evaluate metagenomic techniques for their effectiveness in characterizing microbial communities in various soil types subjected to altered climatic conditions. Through a combination of 16S rRNA gene sequencing and shotgun metagenomic sequencing, we uncovered nuanced distinctions in microbial diversity metrics across different sampling sites. Our findings reveal that shotgun sequencing significantly outperformed 16S rRNA sequencing in capturing rare taxa, leading to a more comprehensive understanding of microbial resilience patterns. Notably, we observed a 35% increase in detected operational taxonomic units (OTUs) and a reduction in sequencing depth requirements by approximately 20%, underscoring the importance of methodological choice in biodiversity assessments.
