Abstract

The precision of CRISPR-Cas9 genome editing is contingent upon the meticulous design of guide RNAs (gRNAs), which is crucial for minimizing off-target mutations. This study introduces a novel multi-objective optimization algorithm to enhance gRNA design, integrating sequence specificity and thermodynamic properties. Through computational simulations and experimental validations, the proposed framework demonstrates superior targeting accuracy and efficiency compared to existing methodologies. These findings suggest a significant advancement in the tailoring of CRISPR-Cas9 tools for precise genome editing applications.
